How immune aging affects cancer immunotherapy
How does immune aging affect the likelihood that cancer immunotherapy will work?
It can clarify whether immune aging explains differences in treatment benefit and whether measuring it could help guide immunotherapy choices.

Where it stands now
Main bottleneck
Determining which measurable features of immune aging reliably predict immunotherapy response, including when they signal better or worse outcomes. The review-only evidence trajectory does not establish a validated predictor.

Since the previous research note: When aging changes the immune neighborhood around a tumor
Previous expectation: Interrupting harmful senescence-related interactions around a tumor might improve checkpoint treatment, provided benefit and safety could be demonstrated in patients.
What this study adds: The current review gives this installment a specific immune-cell focus, including mouse-model accounts of BFAR inhibition restoring aged CD8 T-cell function and enhancing anti-PD-1 effects. Its account of tumor-killing senescence-like T cells also sharpens the distinction between removing cells and restoring their functions.

Since the previous research note: Should cancer treatment remove impaired T cells or restore them?
Previous expectation: A future adjunct to checkpoint treatment might restore useful T-cell activity and improve cancer control with acceptable toxicity.
What this study adds: The March 11 review connects immune aging with clinical variation across cancers, treatment tolerance and the conditions surrounding tumors. Its experimental examples include both senescent-cell clearance and induced tumor senescence as routes toward improved checkpoint response.
